Anzugeben sind die Zytotoxizitaet , die Lebensfaehigkeit , die Koloniezahlen und die Mutantenhaeufigkeit fuer die Pruef- sowie Kontrollkulturen . Ergibt der L5178 / Y TK + / - -Test einen positiven Befund , werden die Kolonien bei mindestens einer Konzentration der Pruefsubstanz ( der hoechsten positiven Konzentration ) sowie bei den Negativ- und Positivkontrollen nach dem Kriterium kleine oder grosse Kolonie ausgewertet . Die molekulare und zellgenetische Beschaffenheit von Mutanten mit Bildung grosser und Mutanten mit Bildung kleiner Kolonien ist gruendlich erforscht ( 23 ) ( 24 ) . Beim TK + / - -Test werden die Kolonien nach dem Kriterium normal wachsende ( grosse ) oder langsam wachsende ( kleine ) Kolonie eingestuft ( 25 ) . Mutantenzellen mit einer erheblichen genetischen Schaedigung weisen eine laengere Generationszeit auf und bilden daher kleine Kolonien . Die Schaedigung kann vom Verlust eines kompletten Gens bis zu karyotypisch erkennbaren Chromosomenaberrationen reichen . Mutanten mit der Bildung kleiner Kolonien treten vor allem bei Chemikalien auf , die starke Chromosomenaberrationen hervorrufen ( 26 ) . Weniger stark betroffene Mutantenzellen wachsen in aehnlichem Tempo wie die Elternzellen und bilden grosse Kolonien .

Data should include cytotoxicity and viability determination , colony counts and mutant frequencies for the treated and control cultures . In the case of a positive response in the L5178Y TK + / - test , colonies are scored using the criteria of small and large colonies on at least one concentration of the test substance ( highest positive concentration ) and on the negative and positive control . The molecular and cytogenetic nature of both large and small colony mutants has been explored in detail ( 23 ) ( 24 ) . In the TK + / - test , colonies are scored using the criteria of normal growth ( large ) and slow growth ( small ) colonies ( 25 ) . Mutant cells that have suffered the most extensive genetic damage have prolonged doubling times and thus form small colonies . This damage typically ranges in scale from the losses of the entire gene to karyotypically visible chromosome aberrations . The induction of small colony mutants has been associated with chemicals that induce gross chromosome aberrations ( 26 ) . Less seriously affected mutant cells grow at rates similar to the parental cells and form large colonies .
