Nach der Applikation der Testsubstanz zur Bestimmung des TER werden 150 l Sulforhodamin B-Farbstoff in 10 % iger ( w / v ) Verduennung in destilliertem Wasser fuer zwei Stunden topisch auf jedes Hautstueck aufgebracht . Die Hautstuecke werden sodann unter fliessendem Wasser von hoechstens Raumtemperatur etwa 10 Sekunden lang mit einem Wasserstrahl gewaschen , um ueberschuessige / ungebundene Farbe zu entfernen . Jedes Hautstueck wird sorgfaeltig vom PTFE-Rohr entfernt und in ein geeignetes verschliessbares Gefaess ( z. B. ein 20-ml-Scintillationsflaeschchen ) mit entionisiertem Wasser ( 8 ml ) gebracht . Die Flaeschchen werden 5 Minuten lang vorsichtig geschuettelt , um ueberschuessige / ungebundene Farbe zu entfernen . Nach Wiederholung des Spuelvorgangs werden die Hautstuecke in Flaeschchen mit 5 ml 30 % igem ( w / v ) Natriumdiodecylsufat ( SDS ) in destilliertem Wasser ueberfuehrt und ueber Nacht bei 60 ° C im Brutschrank aufbewahrt . Nach der Inkubation wird jedes Hautstueck entfernt und verworfen , und die verbleibende Loesung wird 8 Minuten lang bei 21 ° C zentrifugiert ( relative Zentrifugalkraft [ sim ] 175 ) . Eine 1-ml-Probe des oberflaechenaktiven Stoffs wird sodann 1 : 5 ( v / v ) [ d. h. 1 ml + 4 ml ] mit 30 % iger ( w / v ) SDS-Loesung in destilliertem Waser verduennt . Die optische Dichte ( OD ) der Loesung wird bei ca. 565 nm gemessen .

The skin discs are then jet washed with tap water at up to room temperature for approximately 10 seconds to remove any excess / unbound dye . Each skin disc is carefully removed from the PTFE tube and placed in a vial ( e.g. a 20 ml glass scintillation vial ) containing deionised water ( 8 ml ) . The vials are agitated gently for 5 minutes to remove any further excess / unbound dye . This rinsing procedure is then repeated , after which the skin discs are removed and placed into vials containing 5 ml of 30 % ( w / v ) sodium dodecyl sulphate ( SDS ) in distilled water and are incubated overnight at 60 ° C. After incubation , each skin disc is removed and discarded and the remaining solution is centrifuged for 8 minutes at 21 ° C ( relative centrifugal force [ sim ] 175 ) . A 1 ml sample of the supernatant is then diluted 1 in 5 ( v / v ) ( i.e. 1 ml + 4 ml ) with 30 % ( w / v ) SDS in distilled water . The optical density ( OD ) of the solution is measured at approximately 565 nm .
