In this study, the role of GCS in conferring chemoresistance was further investigated with a focus on NOX and implications on oxidative stress. Previously, we had demonstrated that TNFα, a known stimulator of ceramide, elicited oxidative stress by upregulating NOX activity in SH-SY5Y neuroblastoma cells. 19 Similarly, many chemotherapeutics have been shown to induce oxidative stress. 20,21 We therefore hypothesized that increased glucosylceramide production through GCS could interfere with NOX activity as a mechanism critical to chemotherapeutic-resistance. Utilizing pharmacological and molecular techniques, we demonstrated that glucosylceramide potently interfered with agonist- stimulated NOX activity. NOX-mediated ROS production was abolished by blocking the functional assembly of cytosolic and membrane subunits of NOX. We further showed in glioblastoma cells that depleting GCS activity not only augmented NOX activity but improved the efficacy of chemotherapy.

In dieser Studie wurde die Rolle der GCS bei der Verleihung Chemoresistenz weiter mit einem Schwerpunkt auf NOX-und Auswirkungen auf oxidativen Stress untersucht. Zuvor hatten wir gezeigt, dass TNF, einem bekannten Stimulator der Ceramid, hervorgerufen durch oxidativen Stress hochreguliert NOX-Aktivität in SH-SY5Y Neuroblastom-Zellen. 19 Außerdem haben viele Chemotherapeutika wurde gezeigt, dass oxidativer Stress zu induzieren. 20,21 Wir haben daher die Hypothese auf, dass eine erhöhte Produktion durch Glucosylceramid GCS konnte mit NOX-Aktivität als ein Mechanismus entscheidend für chemotherapeutische-Resistenz stören. Mit Hilfe pharmakologischer und molekularer Techniken konnten wir zeigen, dass Glucosylceramid potent gestört NOX-Aktivität Agonisten stimulierten. NOX-vermittelte ROS-Produktion wurde durch die Blockierung der funktionalen Montage von cytosolischen und Membran-Untereinheiten von NOX abgeschafft. Wir zeigten in weiteren Zellen des Glioblastoms, dass GCS abbauende Aktivität nicht nur NOX-Aktivität verstärkt, sondern verbessert die Wirksamkeit der Chemotherapie.
