Nelarabin verursachte histopathologische Veränderungen im zentralen Nervensystem (Vakuolisierung der weißen Substanz und degenerative Veränderungen im Großhirn, Kleinhirn und in der Wirbelsäule) von Affen nach 23-tägiger Behandlung mit Nelarabin bei einer Exposition unterhalb der humantherapeutischen Exposition.

Adverse reactions not observed in clinical studies, but seen in animals at exposure levels similar to clinical exposure levels and with possible relevance to clinical use were as follows: nelarabine caused histopathological changes to the central nervous system (white matter) vacuolation and degenerative changes in cerebrum, cerebellum and spinal cord of monkeys after treatment with nelarabine daily during 23 days, at exposures below the human therapeutic exposure.

Dabei sollen normalerweise die folgenden Bereiche untersucht werden: Vorderhirn, Mitte des Großhirns einschließlich eines Anschnitts durch den Hippokampus, Mittelhirn, Kleinhirn, Brücke, verlängertes Mark, Auge mit Sehnerv und Netzhaut, Rückenmark an den zervikalen und lumbalen Verdickungen, dorsale Wurzelganglien, dorsale und ventrale Nervenwurzelfasern, proximaler Ischiasnerv, proximaler Schienbeinnerv (am Knie) und Wadenmuskel-Äste des Schienbeinnervs.

The areas examined should normally include: the forebrain, the centre of the cerebrum, including a section through the hippocampus, the midbrain, the cerebellum, the pons, the medulla oblongata, the eye with optic nerve and retina, the spinal cord at the cervical and lumbar swellings, the dorsal root ganglia, the dorsal and ventral root fibres, the proximal sciatic nerve, the proximal tibial nerve (at the knee) and the tibial nerve calf muscle branches.
