Reagenz zur Verwendung bei einem Verfahren gemäß Anspruch 1, im Wesentlichen bestehend aus einem Protein von mindestens etwa 25 kDal, gebunden an ein Enzymdonorfragment, das direkt oder durch einen Linker von nicht mehr als etwa 50 Aminosäuren an eine Erkennungssequenz, die durch eine Protease zur proteolytischen Spaltung erkannt wird, gebunden ist, wobei die Erkennungssequenz an eine Oberfläche oder ein Liposom durch einen Linker von nicht mehr als etwa 300 Aminosäuren gebunden ist, wobei das Enzymdonorfragment als Teil des Proteinreagenzes im Wesentlichen nicht in der Lage ist, mit einem Enzymakzeptorfragment unter Bildung eines aktiven Enzyms zu komplexieren, und in der Lage ist, mit einem Enzymakzeptorfragment unter Bildung eines aktiven Enzyms zu komplexieren, wenn die Erkennungssequenz gespalten wird.
