Here, we present a method for applying alginate multilayers to a barium-alginate core without the use of fibrosis-evoking polycations. We report on the factors that influence layer formation and stability and can therefore provide data for full adjustability of the additional layer. Although known for yeast and plant cells, this technique has not previously been demonstrated with mammalian cells or ultra-high viscous alginates. Also, there are no previous studies on the mechanism of layer deposition. Viability of murine insulinoma cells was investigated by live-dead staining and live cell imaging. No hampering effects of the second alginate layer were found. This multi-layer technique therefore has great potential for clinical and in vitro use and is likely to be central in alginate matrix based immunoisolated cell therapy.

Hier stellen wir ein Verfahren zum Aufbringen einer Alginat Multischichten Barium-Alginat Kern ohne Verwendung von Fibrose-evozierende Polykationen. Wir berichten über die Faktoren, die Schichtbildung und Stabilität beeinflussen kann und daher Daten für volle Einstellbarkeit der zusätzlichen Schicht. Obwohl für Hefe-und Pflanzenzellen bekannt ist, hat diese Technik bisher nicht mit Säugerzellen oder ultrahohem viskose Alginate nachgewiesen. Auch gibt es keine früheren Untersuchungen zum Mechanismus der Schichtabscheidung. Lebensfähigkeit der murine Insulinom-Zellen wurde durch Live-dead Färbung und Live Cell Imaging untersucht. Keine Behinderung Wirkungen der zweiten Alginat-Schicht gefunden. Diese Multi-Layer-Technik hat daher großes Potenzial für die klinische und In-vitro-Verwendung und ist wahrscheinlich zentral in Alginat-Matrix immunoisolated Zelltherapie.
