Recovery of the applied micro organism Bacillus subtilis, Candida albicans for TAMC and Aspergillus brasiliensis for TYMC during “Microbial enumeration tests” During performance of the validation the recovery of micro organism Bacillus subtilis for the first test (31.07.2012) is 4% instead ≥ 70%. For Candida albicans and Aspergillus brasiliensis the neutralizer toxicity is -33% resp. -45% instead ± 30%. At the moment the root cause for this is unknown. For example a laboratory error could be the reason (e.g. pipetting mistake of the test germ, incorrect mixture of the sample preparation with the test germs). Therefore this part of the validation shall be repeated with a threefold repetition at the same test date without changes regarding the procedure.

Beispielsweise ein Laborfehler könnte der Grund (zB Pipettieren Fehler der Testkeim, fehlerhafte Mischung der Probenvorbereitung mit den Testkeimen) sein. Daher wird dieser Teil der Validierung ist mit einer dreifachen Wiederholung am gleichen Test Datum ohne Änderungen in Bezug auf das Verfahren wiederholt werden.
