Therapeutische Zusammensetzung, umfassend eine isolierte Zelle, welche ein wirksames Antigen präsentiert, wobei die Zelle stimuliert wird durch die in vitro-Exposition mit einem isolierten löslichen Polypeptidkomplex, welcher ein eine dendritische Zelle bindendes Protein umfasst, das ein Granulozyten-Makrophagen-Kolonie stimulierender Faktor (GM-CSF) ist, welcher kovalent mit einem Polypeptidantigen verknüpft ist, wobei das Polypeptidantigen ausgewählt ist aus der Gruppe bestehend aus einem gewebespezifischen Tumorantigen, wobei das gewebespezifische Tumorantigen charakterisiert ist als (i) Antigene einschließend, welche einem spezifischen Tumortyp gemeinsam sind, und (ii) Antigene ausschließend, welche nur für einen individuellen Tumor spezifisch sind, und einem Onkogenprodukt, ausgewählt aus der Gruppe bestehend aus Her-2, p21RAS und p53, wobei die Zusammensetzung wirksam ist bei der Aktivierung von T-Zellen, um eine multivalente zelluläre Immunantwort gegen das Polypeptidantigen bei einem T-Zell-Aktivierungsgrad zu erzeugen, welcher höher ist als der, welcher durch eine solch ein wirksames Antigen präsentierende Zelle, die durch das Antigen alleine stimuliert wird, erzeugt wird.

A therapeutic composition, comprising an isolated potent antigen presenting cell wherein said cell is stimulated by exposure in vitro to an isolated soluble polypeptide complex comprising a dendritic cell-binding protein being granulocyte macrophage colony stimulating factor (GM-CSF) covalently linked to a polypeptide antigen, said polypeptide antigen selected from the group consisting of a tissue-specific tumor antigen, said tissue-specific tumor antigen characterized as (i) inclusive of antigens that are common to a specific type of tumor and (ii) exclusive of antigens that are specific only to an individual tumor, and an oncogene product, selected from the group consisting of Her-2, p21 RAS and p53, wherein said composition is effective to activate T-cells to produce a multivalent cellular immune response against said polypeptide antigen, at a T-cell activation level higher than that produced by such a potent antigen presenting cell stimulated by said antigen alone.
